The Answer
1- preparation of guaiacum depend on melting of resin upon heating
a- Log of guaiacim is placed horizontally by any support and incision is formed in the middle in the lower surface . Fire is applied in both lateral ends then resin is melt and exudated fron incision and then collected.
b- Log of guaiacim is raised from one end than the other and fire is applied on the raised one then resin is melted and exudated from thewer end
c- By making incision in the trunk of guaiacum ahd then resin is exudated in the form of tears
d- Extract the resin dy shaking with alcohol then pour in water container .. Resin is ppt then separate it and wash with water
2- By making puncturing in the bark in early summer - then in autumn the bark which is saturated with balasm is peeled and boiled in hot H2O or pressed to separate the balasm - then dissolve the balasm in hot alcohol then make filteration and evapourate the filterate at low temp
3- Cape Aloe give green color - Curacao Aloe give reddish brown color - Zinzaber&Socotrine Aloe give yellowish brown color
4- Myrrh powder is shaked with alcohol and alcohol insol. part (Gum Acacia Type) is separated then dissolved in water + Guaiacum Tincture give intense blue color
5- Sumatra benzoin: Contain Benzo resin which composed of a-Ester of cinnamic acid with resinotannol&benzoresinol b-Free cinnamic acid and small amount of benzoic acid c-Traces of vanillin&benzaldehyde
Siam benzoin: Contain Siabezo resin composed of a-Ester of benzoic acid with siaresinotannol%benzoresinol b-Free benzoic acid and traces of cinnamic acid and vanillin

1- a-Normal collection:Olea gum resin is naturally present in schizogenous duct in phloem&bark and may exudated by incision or natural fissures
b-Liquid collection:By injection the plant with phytohormones which absorb water and incease the internal pressure that make releasing for oleo gum resin in less viscous form
2-Sumatra Benzoin : plats crystal due to cinnamic acid & small rod like crystals due to benzoic acid
Siam Benzoin : Long rod like crystals due to benzoic acid
3- A.C: Resinous acid=Gambogic acid - Gambogenic acid - Gambogellic acid - Garcinolic acid(Alpha-Beta-Gama) & Gum of Acacia type with oxidase enzymes
Uses: Cathartic(purgative) ,Hydragogue & Anti-inflamatory ,Analgesic(NSAID) & Cytogenic activity against HEL (Human Embryonic Lung)
4- Triturate the powder with water then make ether extraction then make fillteration - divide the fiterate into two portions 1- evapourate+few drops of HNO3 give Reddish violet color 2- evapourate+vapour of Br2 give Violet color

b-Natural gum :Abnormal product produced by gammosis (conversion of cell wall of certain tissues in to gum due to pathological effect like incision or un favourable conditionmainly occure in cambium & young phloem as in gum acacia or in pith & M.R as in gum tragacanth) - slight levorotatory
16- Skin tendons : are moisted in lime Ca(OH)2 for (Saponification of faty materials - Partial bleaching - Removal of Chondro protein - Prevent putrefaction of protein) then make washing with water
Bones : Are first applied to compaction then boiled with organic solvent like petrolum ether - Bones is dissolved in dil.HCl so that CaCl2 is formed and collagen is separated - Wash with Water - Then boil with water under pressure for extraction of glutin(from collagen) - Then make evapouization of water (30-60 C) Slowly to prevent formation of air bubble or darking of color - Then bleach with SO2
17- By Gambir-Fluorascine test:
Alcoholic Soln. of catechu + 2dps NaOH then Shake with petrolum ether (Pale catechu give rrilliant green fluorescence due to presence of gambir fluorscin -but- Dark catechu not give result -ve)
18- Barbaloin with borax give Aloe emodin Anthranol (Hydrolysis) -but- Barbaloin give with FeCl3/HCl give Aloe emodin(Oxidation)
19- Cape Aloe give green color - Curacao Aloe give reddish brown color - Zinzaber&Socotrine Aloe give yellowish brown color
20- Residue of Mexican Scammony wt is not less than 0.6 gm -but- Residue of Levant Scammony wt is not less than 0.7 gm
21- Ferula Foetida: Give Pink - Reddish purple - Reddish brown (Red variety) As it contain asaresinotannol on oxidation give reddish brown color
Ferula Rubricaulis:Give Yellowish white (Whitw variety) As it free from asaresinotannol
