بس يعني ممكن
ان احنا لما حطينا S زودنا ال polarity بقى more hydrophilic
وكان مكتوب في البنزيل بنسلين انه

فبالتالي لما زادت ال polarity في رقم 3 في السيفالوسبورين بقى كويس بالنسبة للجرام نيجاتيف و اقل اكتيفيتي على الجرام بوزيتيف

-بتغير فى قدرة المضاد الحيوى على الأمتصاص
فتخليه orally active
أما الفاعلية ضد الجرام نيجاتيف بكتريا فبتيجى
من 1- أنه بيحتوى على مجموعة polar فى ال side chain
زى البيبرازين وغيره اللى بيزودوا من قدرة المضاد الحيوى على الدخول فى الجرام نيجاتيف
بكتريا
2-أن المضاد الحيوي يكون بيحتوى على steric sheild بيحميه من البيتا لاكتماز
واللى جاى ده قواعد التعرف على اذا كان السفالوسبورن orally acive

1. First you want to look at the C3 substituent and determine if it is acid labile. If the C3 substituent is acid labile, then the compound is NOT orally active. Currently, the only acid labile group at this position is the acetoxymethyl group. If the C3 substituent is acid stable, procede to step 2.
2. In order for the compound to be orally active, it must have ONE of the following structural features:
-an a-amino group
-a lipid soluble ester
-a vinyl group at the C3 position
-a carboxylic acid present as part of the R1 substituent (It should NOT be directly attached to the alpha-carbon)
3. If the compound has an acid stable group at C# (i.e., does not have an acetoxymethyl group) and has one of the structural features listed in step 2 above, then it is orally active
