السلام عليكم ورحمه الله وبركاته

ان شاء الله ده عملى الصيدلانيات ............ السكشن الثانى


ان شاء الله هنشتغل 2 medicated prescriptions على ال effervescent granules ..........

1st prescription ......... preparation of antispasmodic effervescent granules

طريقه السؤال فى الامتحان كالتالى:

send 20 gm eff. gr. containing 5% liquid extract of Belladonna with wet granulation technique

ده بس اللى بيجى فى السؤال ........ لكن بالنسبه للجزء الثابت فى اى prescription على ال eff. gr. اللى هو:

NaHCO3 510 gm

citric acid 180 gm

tartaric acid 270 gm

sucrose 150 gm

total weight 1110 gm

ده مش بيجى فى السؤال ...... علشان كده لازم نكون حافظينه ان شاء الله .......

نفس طريقه ال calculation لاى prescription :

Required amount = 20 gm

Excess amount = 20 * (25/100) = 5 gm

ال excess amount دى بنضيفها على ال required amount علشان نعوض ال loss
اللى بيحصل اثناء التحضير لان معظم ال ingredients بتكون فى صوره powder
وبالتالى هيحصل loss غصب عننا

وال excess amount دى بتكون ربع الكميه المطلوبه (25%)

so ... Total amount = 20 + 5 = 25 gm

Drug amount = 25 * (5/100) = 1.25 gm = 1.25 ml >>>> liquid

so ..... real amount of the base = 25 - 1.25 = 23.75 gm


Amount of each ingredient


Amount of NaHCO3 = 23.75 * (510/1000 ) = 12.11 gm

Amount of citric acid = 23.75 * (180/1000) = 4.28 gm

Amount of tartaric acid = 23.75 * (270/1000) = 6.41 gm

Amount of sucrose = 23.75 * (150/1000) = 3.56 gm


Method of preparation

نفس ال Method اللى اشتغلنا بيها السكشن اللى فات لكن

الفرق المره دى ان ال Drug هنا liquid مش solid

powder وبالتالى الطريقه هتكون كالتالى :

1. in stainless steel dish ......... evaporate liquid extract of Belladonna on boiling water bath until honey consistency


هناخد كميه ال liquid extract of Belladonna ب cylinder 10 ونحطها فى

stainless steel dish على W.B لحد ما يصبح قوامه viscous مثل قوام العسل

لكن مش يتحرق ( مهم جدا )

دى الخطوه الزياده فى ال prescription دى وباقى الخطوات زى السكشن اللى فات ان شاء الله

2. grinding each powder separately in porcelain mortar

3. mix the 4 powders geometrically

ملحوظه : واحنا بنجهز ال powders ناخد بالنا من ال stainless steel dish اللى

على ال W.B مش ننساه علشان ال liquid extract of Belladonna مش يتحرق (مهم)

كده يبقى عندنا ال powders جاهزه فى ال porcelain mortar وال drug جاهز فى ال stainless steel dish

4.transfer the powder from mortar to the dish ......... then triturate by pestle .........until..... homogenous color is obtained

ملحوظه : فى الخطوه دى لازم نقلب بال pestle مش باى حاجه تانيه ولازم نقلب كويس

جدا علشان يدينا homogenous color والبودر دى بيكون لونها بيج

5. فى t.t. فيها 95%ethanol ننزله dropwise على ال pw. ونقلبه بايدنا مش باى حاجه تانيه لحد ما يدى doughy mass عجينه شبه الاسفنج كده اقدر امسكها فى ايدى

والمفروض مش ننزل كميه كبيره من الكحول علشان ميبقاش زى الميه (مهم جدا )

6.ال doughy mass دى هنضعها على seive no.10 ونحطه على paper

ونحركها شويه علشان مش تعمل lumps (كتل)

لازم ال seive يكون نظيف ونختار اشد جزء فيه ونضغط على ال mass فى اتجاه عمودى واحده واحده مش قوى علشان تدينا eff. gr. كويسه (مهم)


كده اللى هيخرج من ال seive ده هو ال eff. granules نتركها few minutes مش كتير علشان يحصلها drying


7.الخطوه الاخيره والمهمه جدا اننا بعد كده نضع ال eff. granules على طول فى well closed container علشان ال moisture وبكده يبقى الشغل خلص

Evaluation


فى measuring cylinder نضع 10ml water "وممكن نزودها شويه" ونضيف 0.25gm من ال eff. gr. عليها طبعا هتدى effervescence

المطلوب مننا اننا نحدد 4 نقاط هى :

1. onset of effervescence in seconds

وده غالبا بيكون immediately وده دليل ان ال eff. gr. كويسه

2. duration of effervescence in seconds

3. volume of eff

4.clarity of the solution

وده له حالتين : 1.لو كان ال drug ....... soluble in water تبقى clear

2.لكن لو كان drug ....... insoluble in waterتبقى turbid

والاربع نقاط دول بنكتبهم فى جدول فى نهايه الشغل ان شاء الله

2nd prescription .......... preparation of Anti-rheumatic eff. gr


طريقه السؤال فى الامتحان كالتالى :

send 20 gm eff. gr. containing 25% Anti-rheumatic mixture formed of
(sod.salicylate = 2 gm and peppermint oil = 0.1 gm )

الفكره هنا ان ال drug اللى عندى عباره عن حاجتين مع بعض مش حاجه واحده بس

زى ال prescriptions اللى فاتت

بنفس الطريقه اللى بنمشى بيها على طول:


Required amount = 20 gm

Excess amount = 20 * (25/100) = 5 gm

so ... Total amount = 20 + 5 = 25 gm

Drug amount = 25 * (25/100) = 6.25 gm

so ..... real amount of the base = 25 - 6.25 = 18.75 gm

Amount of citric acid = 18.75 * (180/1000) = 3.38 gm

بالنسبه بقى للجزء الزياده هنا :


عندنا amount of drug = 6.25 gm هنقسمهم على الحاجتين اللى بيتكون منهم ال drug كالتالى :


amount of sod.salicylate = 6.25 * ( 2 / 2.1 ) = 5.95 gm

amount of peppermint oil = 6.25 * ( .1 / 2.1 ) = 0.3 gm = 0.3 ml

ملحوظه : بنجيب كميه كل واحد عن طريق :

نضرب ( كميته فى ال drug / مجموعهم ال2 ) * كميه ال drug الكليه فى ال prescription


بالنسبه ل amount of peppermint oil = 0.3 ml هاخدها ازاى ؟؟

هناخدها بال dropper بحيث ان كل ......... one drop = 0.05 ml

اذآ 0.3 على 0.05 يدينا عدد ال drops اللى هنحطها = 6 drops

هى بالظبط ال general steps لكن فيه خطوه بسيطه زياده :

1.grinding separately ( وال sod. salicylate ده عباره عن
solid powder وبالتالى هنطحنه عادى مفيش مشكله )


2.geometric mixing in porcelain mortar

3. wetting by 95% ethanol until doughy mass

الخطوه الزياده :

بعد ما تتكون ال doughy mass ونقدر نمسكها ........ وهى فى ايدنا نحط عليها ال

peppermint oil بال dropper قبل ما نعمل خطوه ال sieving على طول

4. sieving

5. drying


ونكمل بنفس الطريقه وفى الاخر برده نعمل ال evaluation ان شاء الله .......

Important precautions for preparation of any prescription ....


1. لازم كل الادوات تكون dry

2.نبعد عن ال W.B واحنا شغالين بنجهز ال pw

3. well grinding of powders especially citric acid , sucrose (crystalline
